Protein carbonyls are formed by a variety of oxidative mechanisms and can be used as an index of oxidative injury. The protein carbonyl ELISA Assay kit is recommended as a means to measure generalized oxidation of proteins in plasma, serum, cell lysates, tissue homogenates, BAL, CNS and other biological sample types.

The historical method for protein carbonyl analysis was a colorimetric procedure based on the extinction coefficient of a hydrazone compound produced by the reaction of 2,4-dinitrophenylhydrazine (DNPH) with protein carbonyls. In contrast, the Biocell PC Assay utilizes an antibody to directly detect protein carbonyl levels after derivatization and quantify them through comparison to a set of protein carbonyl standards. This method, first published by Hendrikje Buss, Christine Winterbourn et al in 1997 and later commercialized by Zenith Technology Corporation (Zentech) has become the most widely cited assay for protein carbonyl measurement. In recent years, Biocell Corporation has continued to refine the method and carefully optimize it to provide the following benefits compared to other commercially available assays of this type.
