Summary

Protein carbonyls are formed through oxidation of proteins by a variety of mechanisms. They are sensitive markers of oxidative injury. The customary way of analysing protein carbonyls is a colorimetric procedure that measures binding of dinitrophenylhydrazine (DNP). This test is an ELISA format using an anti-DNP antibody developed by: I. Hendrikje Buss, Christine C. Winterbourn and coworkers at the Free Radical Research Group, Christchurch School of Medicine (University of Otago), Christchurch, New Zealand.

Key Benefits:

  • Measures carbonyls quantitatively
  • Requires microgram amounts of protein
  • Has a very low non-specific background
  • Is highly sensitive for analysing clinical samples
  • Correlates directly with the classical colorimetric assay
  • Is easier to use, less labour-intensive and handles more samples per day than the colorimetic assay
  • Strip-well plates allow partial use of the kit for smaller runs

Catalog Number: NWK-PCK01
Methodology: 96 Well ELISA
Sample Requirements: Biological fluids such as plasma, serum, bronchoalveolar lavage fluid and cerebrospinal fluid, to cell extracts and to other soluble protein samples.
Specificity: Protein carbonyls
Sensitivity: 0.05 ng/mL
Expected Values: The protein carbonyl content of plasma from most healthy human adults, as determined by ELISA, is below 0.1 nmol/mg. It is elevated in some disease states (please refer to the published literature for details).
Tests per Kit: 96 Wells...27 samples in triplicate, 41 in duplicate
Storage and Stability: Six months from the date of manufacture when stored as specified
